**Purpose of Use:** This kit is designed for the quantitative determination of Type I procollagen C-terminal peptide (CICP) in human serum, plasma, and other related biological samples. It provides a reliable method for assessing collagen synthesis markers, which can be useful in diagnosing and monitoring conditions such as liver fibrosis, osteoporosis, and other connective tissue disorders.
**Experimental Principle:** The assay is based on the double-antibody sandwich ELISA method. A microplate is pre-coated with a specific monoclonal antibody against CICP. After adding the sample, the CICP present in the sample binds to the immobilized antibody. An HRP-conjugated secondary antibody is then added, forming a complex of antibody-antigen-enzyme-labeled antibody. Following a thorough wash step, TMB substrate is added. Under the catalytic action of HRP, TMB turns blue and then changes to yellow upon addition of the stop solution. The intensity of the color is directly proportional to the concentration of CICP in the sample. The absorbance is measured at 450 nm using a microplate reader, and the CICP concentration is determined by comparing the sample OD values to a standard curve.
