The research team led by Professor Qiao Mingqiang and Dr. Yang Wen from the School of Life Sciences at Nankai University has made a significant breakthrough in the study of natural biological preservatives. For the first time, they have successfully revealed the cutting mechanism of the C-terminus of the NisP leader protein in *Streptococcus mutans*. This discovery provides a solid theoretical foundation for the engineering production of natural antibacterial peptides. The findings were recently published in the prestigious international journal *Acta Crystallographica Section D*.
Currently, chemical preservatives such as sodium nitrite are widely used in food processing, but they often come with potential health risks. Developing safe, non-toxic, and effective natural preservatives is crucial for improving food safety. Nisin, a naturally occurring antimicrobial peptide, is considered one of the most promising options. It helps reduce the sterilization temperature during food processing, preserving the nutritional value and color of food. Moreover, it can be broken down by human enzymes, making it highly biocompatible. To date, Nisin is the only natural preservative approved by the U.S. Food and Drug Administration (FDA) for use in food.
Despite its benefits, the high production cost of Nisin has limited its widespread application in the food industry. To address this challenge, the Molecular Microbiology and Microbial Engineering Laboratory (3M Lab) led by Professor Qiao Mingqiang and the Structural Biology Laboratory led by Dr. Yang Wen conducted a detailed analysis of the crystal structure and metabolic pathway of NisP. They discovered that the C-terminus of the NisP protein contains a self-cleavage site, which plays a key role in the processing of the peptide.
Professor Qiao Mingqiang emphasized, "This self-cleavage site opens up new possibilities for the efficient and low-cost production of natural biopreservatives. It brings us closer to replacing harmful chemical preservatives with safer alternatives. We are committed to furthering our research to ensure that natural biopreservatives become a reliable solution for food safety challenges in the future."
